چکیده

To introduce temporal control in genetic experiments targeting the endothelium, we established a mouse line expressing tamoxifen-inducible Cre-recombinase (Cre-ER) under the regulation of the vascular endothelial cadherin promoter (VECad). Specificity and efficiency of Cre activity was documented by crossing VECad-Cre-ER with the ROSA26R reporter mouse, in which a floxed-stop cassette has been placed upstream of the -galactosidase gene. We found that tamoxifen specifically induced widespread recombination in the endothelium of embryonic, neonatal, and adult tissues. Recombination was also documented in tumor-associated vascular beds and in postnatal angiogenesis assays. Furthermore, injection of tamoxifen in adult animals resulted in negligible excision (lower than 0.4%) in the hematopoietic lineage. The VECad-Cre-ER mouse is likely to be a valuable tool to study the function of genes involved in vascular development, homeostasis, and in complex processes involving neoangiogenesis, such as tumor growth. Inducible Cre mice.

The Cre/lox site-specific recombination system has emerged as an important tool for the generation of conditional somatic mouse mutants. This method allows one to control gene activity in space and time in almost any tissue of the mouse, thus opening new avenues for studying gene function and for establishing sophisticated animal models of human diseases. A major technical advance in terms of i...

Pitx3-CreER mice showing restricted Cre expression in developing ocular lens and skeletal muscles.

To generate temporally controlled inactivation or activation of interested genes in Pitx3-expressing cells, the tamoxifen-inducible form of Cre, CreER(T2), was inserted into the Pitx3 locus of a mouse BAC clone. Following a single dose of tamoxifen, Cre activity in Pitx3-CreER(T2) transgenic mice was observed in the ocular lens and skeletal muscles but not in the central nervous system at vario...

Selectable Marker Gene Removal and Expression of Transgene by Inducible Promoter Containing FFDD Cis-Acting elements in Transgenic plants

Abstract Background: Selectable marker gene (SMG) systems are critical for generation of transgenic crops. Transgenic crop production Background: Selectable marker gene (SMG) systems are critical for generation of transgenic crops. Transgenic crop production without using SMG is not economically feasible. However, SMGs are non-essential once an intact transgenic plant has been established. Eli...

Impaired fertility and spermiogenetic disorders with loss of cell adhesion in male mice expressing an interfering Rap1 mutant.

The guanosine trisphosphatase Rap1 serves as a critical player in signal transduction, somatic cell proliferation and differentiation, and cell-cell adhesion by acting through distinct mechanisms. During mouse spermiogenesis, Rap1 is activated and forms a signaling complex with its effector, the serine-threonine kinase B-Raf. To investigate the functional role of Rap1 in male germ cell differen...

Expression Pattern of the Synthetic Pathogen-Inducible Promoter (SynP-FF) in the Transgenic Canola in Response to Sclerotinia sclerotiorum

Sclerotinia sclerotiorum is a phytopathogenic fungus which causes serious yield losses in canola. A pathogen inducible-promoter can facilitate the production of Sclerotinia-resistant transgenic canola plants. Inthis study, the “gain of function approach” was adopted for the construction of a pathogen-inducible promoter. The synthetic promoter technique was used, which involved the in...
